ABSTRACT

Foot-and-mouth disease (FMD) is an acute, severe, and highly contagious infectious disease caused by foot-and-mouth disease virus (FMDV), which seriously endangers the development of animal husbandry. The inactivated FMD vaccine is the main product for the prevention and control of FMD, which has been successfully applied to control the pandemic and outbreak of FMD. However, the inactivated FMD vaccine also has problems, such as the instability of antigen, the risk of spread of the virus due to incomplete inactivation during vaccine production, and the high cost of production. Compared with traditional microbial and animal bioreactors, production of antigens in plants through transgenic technology has some advantages including low cost, safety, convenience, and easy storage and transportation. Moreover, since antigens produced from plants can be directly used as edible vaccines, no complex processes of protein extraction and purification are required. But, there are some problems for the production of antigens in plants, which include low expression level and poor controllability. Thus, expressing the antigens of FMDV in plants may be an alternative mean for production of FMD vaccine, which has certain advantages but still need to be continuously optimized. Here we review the main strategies for expressing active proteins in plants, as well as the research progress on the expression of FMDV antigens in plants. We also discuss the current problems and challenges encountered, with the aim to facilitate related research.

El virus de la fiebre aftosa es un patógeno altamente infeccioso y contagioso. Recientemente, el topotipo VII, linaje Lib-12 del serotipo SAT2 se describió en brotes en Egipto durante 2018. La vacunación es una forma eficaz de controlar y combatir los brotes del virus de la fiebre aftosa, especialmente en áreas endémicas como Egipto. El presente estudio tuvo como objetivo evaluar la eficacia de la vacuna contra la fiebre aftosa que se produce actualmente, frente a la cepa de campo recientemente aislada del virus de la fiebre aftosa SAT2 topotipo VII, linaje Lib-12 (SAT2 Libia), mediante la aplicación de estudios in vitro e in vivo. Se inocularon en terneros, dos lotes de la vacuna actual contra el virus de la fiebre aftosa. A los 28 días posteriores a la vacunación, se recolectaron muestras de suero y se analizaron contra el virus de la fiebre aftosa SAT2 Libia adaptado a cultivo de tejidos y SAT2/EGY/2/2012 utilizando una prueba de neutralización viral para determinar la relación serológica (valor r1). El ensayo de reto en terneros vacunados se llevó a cabo empleando una cepa virulenta de la fiebre aftosa SAT2 Libia. Se encontró que los títulos de anticuerpos neutralizantes inducidos por los dos lotes de vacuna (1 y 2) y los de animales no vacunados, fueron 0,48, 0,39 y 0,15 log10 DICT50/mL, respectivamente, mientras que la prueba reveló valores de protección de 20 por ciento, 0 por ciento y 0 por ciento, respectivamente. Además, los valores de r1 fueron 0,195 y 0,186 para los lotes de vacuna (1 y 2), respectivamente. Se llegó a la conclusión de que los lotes de vacunas locales comerciales inactivadas disponibles actualmente (SAT2 SAT2/EGY/2/2012) no protegen a los terneros contra el virus circulante de la fiebre aftosa SAT2 topotipo VII, linaje Lib-12 que se aisló recientemente, por lo que es recomendable actualizar las vacunas existentes con la cepa aislada actualmente(AU)


Foot and mouth disease virus is a highly infectious and contagious pathogen. Recently the topotype VII, Lib‐12 lineage of serotype SAT2 was reported through outbreaks in Egypt during 2018. Vaccination is an effective way to control and combat the foot and mouth disease virus outbreaks especially in endemic areas like Egypt. The present study was aimed to evaluate the efficacy of the current produced foot and mouth disease vaccine, against the recently isolated field strain foot and mouth disease virus SAT2 topotype VII, Lib-12 lineage (SAT2 Libya), by applying in vitro and in vivo studies. Two batches of the current foot and mouth disease virus vaccine were inoculated in calves. At the 28th day post-vaccination serum samples were collected and tested against tissue culture adapted foot and mouth disease virus SAT2 Libya and SAT2/EGY/2/2012 using virus neutralization test to determine serological relationship (r1-value). The challenge test for vaccinated calves was carried out against the virulent foot and mouth disease virus SAT2 Libya. It was found that neutralizing antibody titers induced by the two vaccine batches (1 and 2) and those in unvaccinated animals were 0.48, 0.39 and 0.15 log10 TCID50/mL, respectively, while the challenge revealed protection values of 20 percent, 0 percent and 0 percent, respectively. Furthermore, the r1 values were 0.195 and 0.186 for vaccine batches (1 and 2), respectively. It was concluded that the available local commercial inactivated foot and mouth disease virus vaccine batches (SAT2 SAT2/EGY/2/2012) are unable to protect calves against the current circulating foot and mouth disease virus field isolate SAT2 topotype VII, Lib-12 lineage, thus it is highly recommended to update the existing vaccines with the present isolated strain(AU)

ABSTRACT

In order to construct a recombinant replication deficient human type 5 adenovirus (Ad5) expressing a foot-and-mouth disease virus (FMDV) capsid protein, specific primers for P12A and 3B3C genes of FMDV-OZK93 were synthesized. The P12A and 3B3C genes were then amplified and connected by fusion PCR, and a recombinant shuttle plasmid pDC316-mCMV-EGFP-P12A3B3C expressing the FMDV-OZK93 capsid protein precursor P12A and 3B3C protease were obtained by inserting the P12A3B3C gene into the pDC316-mCMV-EGFP plasmid. The recombinant adenovirus rAdv-P12A3B3C-OZK93 was subsequently packaged, characterized and amplified using AdMaxTM adenovirus packaging system, and the expression was verified by infecting human embryonic kidney cell HEK-293. The humoral and cellular immunity levels of well-expressed and purified recombinant adenovirus immunized mice were evaluated. The results showed that rAdv-P12A3B3C-OZK93 could be stably passaged and the maximum virus titer reached 1×109.1 TCID50/mL. Western blotting and indirect immunofluorescence showed that rAdv-P12A3B3C-OZK93 expressed the FMDV-specific proteins P12A and VP1 in HEK-293 cells. In addition, the PK cell infection experiment confirmed that rAdv-P12A3B3C-OZK93 could infect porcine cells, which is essential for vaccination in pigs. Comparing with the inactivated vaccine group, the recombinant adenovirus could induce higher FMDV-specific IgG antibodies, γ-IFN and IL-10. This indicates that the recombinant adenovirus has good immunity for animal, which is very important for the subsequent development of foot-and-mouth disease vaccine.

ABSTRACT

The stability of virus-like particles (VLPs) is currently the main factor affecting the quality of foot-and-mouth disease VLPs vaccines. In order to further improve the quality of the VLPs vaccine of foot-and-mouth disease (FMD), three amino acid modification sites were designed and screened through kinetic analysis software, based on the three-dimensional structure of FMDV. The three mutant recombinant plasmids were successfully prepared by the point mutation kit, transformed into Escherichia coli strain BL21 and expressed in vitro. After purification by Ni ion chromatography column, SDS-PAGE proved that the three amino acid mutations did not affect the expression of the target protein. The results of the stability study of three FMD mutant VLPs obtained by in vitro assembly show that the introduction of internal hydrophobic side chain amino acids made the morphology of VLPs more uniform (N4017W), and their stability was significantly improved compared to the other two VLPs. The internal hydrophobic force of the capsid contributes to the formation of VLPs and helps to maintain the stability of the capsid, providing new experimental ideas for improving the quality of VLPs vaccines, and helping to promote the development of VLPs vaccines.

ABSTRACT

Foot-and-mouth disease represents an important barrier to the international commerce of animal products, potentially associated with significant economic losses. The systematic vaccination of bovines and buffaloes was fundamental for the eradication of this disease; however, the use of vaccines can lead to reactions at the application site. The objectives of this study were to evaluate the effects of the vaccination protocol to the production of dairy cows and to observe the occurrence of vaccinal reactions in the animals. At one property located in the municipality of Salvador do Sul, Rio Grande do Sul, 270 dairy cows were vaccinated against foot-and-mouth disease in May 2019. The vaccine was administered via a subcutaneous application using disposable syringes and needles for each animal. Inspection of the animals was performed before and 20 days after the vaccination to verify the presence of reactions to the vaccine. The study's sample was set by convenience, including 203 lactating animals with or without bovine somatotropin (BST) administration during the data collection period, which was limited to 20 days before and 20 days after the vaccination. Milk production data was obtained through SmartDairy® HerdMetrix™ software, tabulated in electronic spreadsheets using Microsoft Excel® and processed using the program SAS®, considering a 5% significance level for mixed model statistical analysis. A total of 160 animals (78.82%) presented local lesions at the application site, even when the recommended vaccination practices were followed, suggesting that the high reaction power was provoked by the vaccinal components. In regards to milk production, a statistically significant (p<0.05) decrease of 0.30kg of milk per animal/day was observed in the average daily production in the 20 days post-vaccination. These results demonstrate the local and systemic effects caused by the foot-and-mouth disease vaccine, evidenced by reduced levels of milk production and the occurrence of vaccine reactions, implying significant economic losses.(AU)


A febre aftosa representa uma importante barreira no comércio internacional de produtos de origem animal, podendo acarretar em significativas perdas econômicas. A vacinação sistemática de bovinos e bubalinos foi fundamental para a erradicação da doença. No entanto, a utilização de vacinas pode causar reações no local da aplicação. O objetivo deste trabalho foi avaliar os efeitos da vacina em bovinos leiteiros e observar a incidência de reações vacinais no local de aplicação. O estudo foi realizado numa propriedade leiteira do município de Salvador do Sul, Rio Grande do Sul, onde foram vacinados 270 bovinos contra febre aftosa no mês de maio de 2019. A vacina foi administrada por via subcutânea, com seringas e agulhas descartáveis para cada animal. Foi realizada inspeção dos animais antes da vacinação e 21 dias após a vacinação, para verificar a presença de reações vacinais. A amostra foi definida por conveniência, incluindo 203 vacas em lactação com ou sem administração de somatotropina bovina (BST) durante o período de coleta de dados, que l foi de 20 dias antes e 20 dias após a vacinação. Estes dados de produção de leite foram obtidos através do software SmartDairy® HerdMetrix™, tabulados em planilhas eletrônicas do Microsoft Excel® e processados usando o programa SAS®, considerando 5% de nível de significância para uma análise estatística modelo misto. Foi observado que 160 (78,82%) vacas apresentaram lesões no local de aplicação, mesmo quando a aplicação era realizada de acordo com as boas práticas de vacinação, o que indica o alto poder de reação provocada pelos componentes da vacina. Em relação à produção de leite, observou-se uma redução significativa (p<0,05) na produção média diária de 0,30kg de leite por animal/dia nos 21 dias após a vacinação. Esses resultados demonstram os efeitos locais e sistêmicos provocados pela vacina da febre aftosa, evidenciados pela redução na produção de leite e pela incidência de reações vacinais, o que implica em significativas perdas econômicas.(AU)

ABSTRACT

Antigenic purity is important for quality control of the foot-and-mouth (FMD) whole virus inactivated vaccine. The recommended method for evaluation the antigenic purity of FMD vaccine is to check the serum conversion to non-structural protein (NSP) 3AB antibody after 2 to 3 times inoculation of animals with inactivated vaccine. In this study, we developed a quantitative ELISA to detect the amount of residual 3AB in vaccine antigen, to provide a reference to evaluate the antigenic purity of FMD vaccine. Monoclonal antibody (Mab) of NSP 3A and HRP-conjugated Mab of NSP 3B were used to establish a sandwich ELISA to quantify the NSP 3AB in vaccine antigen of FMD. Purified NSP 3AB expressed in Escherichia coli was serially diluted and detected to draw the standard curve. The detectable limit was determined to be the lowest concentration of standard where the ratio of its OD value to OD blank well was not less than 2.0. Results: The OD value was linearly corelated with the concentration of 3AB protein within the range between 4.7 and 600 ng/mL. The correlation coefficient R² is greater than 0.99, and the lowest detectable limit is 4.7 ng/mL. The amount of 3AB protein in non-purified inactivated virus antigen was detected between 9.3 and 200 ng/mL depending on the 12 different virus strains, whereas the amount of 3AB in purified virus antigen was below the lowest detectable limit. The amount of 3AB in 9 batches of commercial FMD vaccine antigens was between 9.0 and 74 ng/mL, whereas it was below the detectable limit in other 24 batches of commercial vaccine antigens. Conclusion: the sandwich ELISA established in this study is specific and sensitive to detect the content of 3AB protein in vaccine antigen of FMD, which will be a useful method for evaluation of the antigenic purity and quality control of FMD inactivated vaccine.

ABSTRACT

Cases of compressive myelopathy syndrome associated with post vaccinal pyogranulomas were diagnosed post mortem in three cows from a farm in Minas Gerais state, Brazil. These cows presented ataxia and bilateral paresis of the pelvic limbs, which evolved to paralysis, and sternal recumbence. On necropsy, locally extensive areas of the longissimus dorsi muscle were replaced by pyogranulomas supported by moderate amounts of fibrous connective tissue. On the cut surface, some nodules contained yellowish and viscous fluid (purulent exudate) or whitish fluid (interpreted as the oily adjuvant of a vaccine). In the spinal canal of the subjacent vertebrae, compressing the spinal cord, were pyogranulomas identical to those described in the skeletal muscle. Histologically, the pyogranulomas were composed of a central clear vacuole (consistent with the space left by the oil adjuvant droplets), surrounded by neutrophils and, more externally, by large numbers of epithelioid macrophages and fewer multinucleated giant cells. In the white matter of the spinal cord were numerous well-defined, clear vacuoles (Wallerian degeneration). The association of the clinical history and pathological findings allowed the diagnosis of compressive myelopathy associated with pyogranulomatous reaction to the oily adjuvant of the foot-and-mouth disease vaccine, in this case, due to its inadequate application.(AU)


São descritos casos de síndrome de compressão medular, associada a granulomas pós-vacinais, em bovinos Nelore, provenientes de uma propriedade em Minas Gerais. Esses bovinos apresentavam ataxia e paresia bilateral dos membros pélvicos, que evoluiu para paralisia e decúbito esternal. Na necropsia, áreas focalmente extensas da musculatura na região torácica dorsal (músculo longissimus dorsi) eram substituídas por numerosos piogranulomas, separados por tecido brancacento e firme (tecido conjuntivo fibroso). Ao corte, alguns nódulos continham material amarelado e viscoso (exsudato purulento) ou material esbranquiçado e fluido (sugestivo de adjuvante de vacina). No canal medular das vértebras subjacentes, havia granulomas idênticos aos observados no tecido muscular. Histologicamente, os piogranulomas continham, no centro, vacúolo, bem delimitado e arredondado (consistente com o espaço deixado pela gotícula de lipídio do adjuvante), circundado por variável quantidade de neutrófilos degenerados e íntegros e, mais externamente, por numerosos macrófagos epitelioides e algumas células gigantes multinucleadas. Nas áreas da medula espinhal, circundadas pelos granulomas, numerosos vacúolos, bem definidos, eram observados na substância branca (degeneração walleriana). A associação do histórico clínico e de achados patológicos permitiu o diagnóstico de mielopatia compressiva associada à reação granulomatosa ao adjuvante oleoso da vacina contra febre aftosa, no caso, induzida pela aplicação inadequada da vacina.(AU)

ABSTRACT

Para avaliar a incidência e estimar as perdas econômicas decorrentes de lesões nodulares características de reação vacinal contra febre aftosa foi acompanhado o abate de 72.000 bovinos no Mato Grosso do Sul, de maio a junho de 2018. O estudo foi realizado com base na avaliação macroscópica das lesões, que resultaram em uma incidência/mês média de 99,0% no rebanho abatido. O peso médio de descarte devido ao abscesso foi de 1,56kg/carcaça e a perda estimada foi de 673.920kg/ano ou 44.928 arrobas/ano. O preço médio de venda da carne foi de R$ 11,65/kg, sendo estimada perda de R$ 7.851.168,00/ano (US$ 2.116.117,79/ano). A elevada incidência de lesões nodulares características de reação à vacina contra febre aftosa nos bovinos abatidos no Mato Grosso do Sul representa um problema sanitário e econômico relevante na cadeia da carne.

ABSTRACT

Routine and emergency vaccination of small ruminants against foot-and-mouth disease (FMD) is mandatory in many endemic countries, yet data on the field effectiveness of the vaccines used is scarce. We conducted an investigation of a serotype O FMD outbreak that took place in a sheep and goat pen, and estimated the effectiveness of various routine vaccination statuses. We also evaluated the protection provided by colostrum administration and emergency vaccination. Animals which were routinely vaccinated twice were not clinically affected while disease incidence was observed among animals routinely vaccinated only once (p = 0.004 according to a two-sided Fisher's exact test). In groups vaccinated only once, there was a significant association between the average time that elapsed since last vaccination and the disease incidence (n = 5; Spearman correlation coefficient: r(s) = 1.0, p < 0.01). In addition, non-vaccinated lambs fed colostrum from dams vaccinated more than 2 months before parturition had a mortality rate of 33%. Administration of emergency vaccination 2 days after the occurrence of the index case was the probable reason for the rapid blocking of the FMD spread within 6 days from its onset in the pen.

ABSTRACT

Lesões exacerbadas à vacina contra a febre aftosa foram observadas em 1.815 de um total de 5.000 bovinos abatidos em frigorífico com inspeção federal. Essas lesões resultaram na condenação de parte das carcaças de acordo com o Regulamento da Inspeção Industrial e Sanitária de Produtos de Origem Animal. Os bovinos haviam sido vacinados em maio de 2012 e abatidos em setembro do mesmo ano e divididos em Lotes 1 e 2 constituídos respectivamente por 1.500 e 315 bovinos. As lesões de reação vacinal eram caracterizadas por nódulos protuberantes, circunscritos, bem delimitados, firmes e de tamanhos variáveis, distribuídos multifocalmente na musculatura do aspecto lateral do pescoço; ao corte apresentavam exsudato purulento. Amostras da lesão de 28 bovinos foram avaliadas histologicamente e os principais achados foram piogranulomas com espaços claros circulares de tamanhos variados no centro, circundados por intenso infiltrado de neutrófilos íntegros e necróticos, circundados por macrófagos epitelioides com citoplasma vacuolizado, ocasionais células gigantes multinucleadas e, mais externamente, por abundante tecido conjuntivo em meio observavam-se linfócitos e plasmócitos. Culturas bacterianas realizadas em amostras do exsudato da lesão de seis bovinos resultaram negativas. Quando as carcaças afetadas foram submetidas ao toalete foi retirado em média 1,8 e 2,0 kg de músculo da área afetada, respectivamente dos Lotes 1 e 2. O prejuízo econômico da propriedade de origem dos bovinos afetados foi de R$ 20.424,00, considerando o preço pago pela arroba do boi no mês e ano da ocorrência. Esses valores à época seriam suficientes para adquirir 29,17 bezerros desmamados para engorda. Se considerarmos 5.000 bovinos vacinados forem considerados como população sob risco o coeficiente de morbidade seria de 0,36%. Os resultados deste estudo demonstram que perdas por reação vacinal, mesmo quando não provocam sinais clínicos marcantes, podem ocasionar importantes prejuízos econômicos...


Lesions caused by excessive reaction to foot and mouth vaccine were observed in 1,815 out of 5,000 cattle slaughtered in a slaughterhouse under federal meat inspection. Such lesions resulted in condemnation of the affect parts of the carcasses in accordance with the Brazilian Federal Meat Inspection Regulations. The affected cattle had been vaccinated on May 2012 and then slaughtered in September of the same year. They were allotted to two groups with 1,500 (Groups 1) and 315 cattle (Group 2). Post vaccination lesions were characterized as salient, firm variable sized nodules multifocally distributed in the musculature of the lateral neck. Pus oozed at the cut surface of these nodules. Samples of 28 cattle were submitted for histopathological examination; main findings were pyogranulomas with central variable-sized clear spaces surrounded by marked infiltrate of viable and dead neutrophils, which were surrounded by epithelioid macrophages with vacuolated cytoplasm and occasional multinucleated giant cell. This inflammatory reaction was walled by abundant connective tissue infiltrated by lymphocytes and plasma cells. Samples of the lesion of six cattle were submitted to bacteriological culture with negative results. When affected carcasses went through dressing, there was an average of 1.8 and 2.0 kg of muscle tissue cut off from the carcasses respectively of Group 1 and 2. Economic losses in the farm of origin of the affected cattle was R$ 20,424.00, considering the price paid by kg of cattle for slaughter at the time of the occurrence. This amount at that time was enough to buy 29.17 weaned calves for fattening. If one considers theses 5,000 cattle vaccinated on the farm as the population at risk, the morbidity rate would be 0.36%. These results indicate that the losses due to vaccine reaction, even with no overt clinical signs, may cause important economic losses...

ABSTRACT

El presente estudio evaluó el efecto de la masa antigénica y de diferentes métodos de purificación y concentración del virus de Fiebre Aftosa en la inducción de respuesta de anticuerpos específicos contra proteínas asociadas a la cápside (PC) y no asociadas a la cápside (PNC) en bovinos inmunizados con vacuna oleosa bivalente (A24 Cruzeiro y O1 Campos). Se formularon cuatro vacunas con diferente carga viral por dosis (Vacuna 1, 16.9; Vacuna 2, 8.8; Vacuna 3, 17.9; y Vacuna 4, 7.7 ug/dosis). Se inmunizaron 32 bovinos de 12 a 24 meses de edad (ocho por cada vacuna) a los días 0 y 30. La respuesta serológica contra PC fue evaluada con la prueba de ELISA CFL en términos de Expectativa de Protección Porcentual (EPP) al día 30 y la reactividad a PNC se determinó con el sistema ELISA-I 3ABC/EITB al día 60. Las vacunas formuladas con antígenos virales purificados con sales indujeron mayor EPP promedio tanto para virus A24 Cruzeiro (Vacuna 3, 89.8 por ciento; Vacuna 4, 83.4 por ciento) como O1 Campos (Vacuna 3, 92.6 por ciento; Vacuna 4, 82.2 por ciento) en comparación con los antígenos tratados con Polietilen Glicol cuyos resultados de EPP para virus A24 Cruzeiro fueron: Vacuna 1, 80.2por ciento; Vacuna 2, 71.8 por ciento; y para virus O1 Campos: Vacuna 1, 78.1 por ciento; Vacuna 2, 73.7 por ciento. Adicionalmente, un bovino inoculado con la vacuna 3 fue positivo a PNC a los 60 días post vacunación (dpv). En este estudio se encontró que, dependiendo del proceso de concentración y purificación de antígenos, se pueden obtener resultados diferentes así: para los virus tratados con sales, con una baja (vacuna 4) y alta (vacuna 3) carga antigénica, es posible lograr muy buena inmunogenicidad, mientras que con alta carga antigénica se tiene mayor riesgo de inducir reactividad a PNC; y en el caso de los virus tratados con PEG se obtuvo buena protección, sin evidencia de interferencia en la determinación de los animales infectados cuando fueron evaluados por el sistema....

ABSTRACT

A febre aftosa é uma das doenças mais temidas nos rebanhos em todo o mundo. A vacinação tem sido uma arma eficiente no controle da doença, no entanto há preocupações com as vacinas atualmente utilizadas incluindo a necessidade de instalações de alta segurança para a produção dessas vacinas e a falta de um teste de diagnóstico aprovado que faça distinção precisa entre animais vacinados dos infectados. Várias vacinas têm sido testadas contra a febre aftosa e uma dessas utiliza como vetor um vírus defectivo para replicação, derivado do adenovírus humano tipo 5 (Ad5), o qual contém as proteínas que compõe capsídeo do vírus da febre aftosa (P1-2A) e a protease 3C, protegeu completamente suínos contra o desafio de uma cepa homóloga (A12 e A24). Uma vacina com o Ad5-P1-2A+3C proveniente da cepa O1 Campos (Ad5-O1C), no entanto, somente induziu um baixo título de anticorpos neutralizantes específicos em testes de potência vacinal em suínos. O fator estimulante de colônias de granulócitos e macrófagos (GM-CSF) tem sido utilizado com sucesso na formulação de vacinas para estimular a resposta imune contra inúmeras doenças, incluindo HIV, Hepatite C e B. Na tentativa de melhorar a resposta imune específica contra a febre aftosa induzida pelo Ad5-O1C, suínos foram vacinados com Ad5-O1C juntamente com Ad5-GM-CSFporcino. Entretanto nas, condições utilizadas nesse teste, o GM-CSF suíno não melhorou a resposta imune do Ad5-O1C e adversamente afetou o nível de proteção de suínos desafiados com o vírus homólogo da febre aftosa.

La cíclica y masiva movilización de terneros puede determinar la recurrencia y persistencia de fiebre aftosa. Por ello, en Sudamérica se promueven esquemas de vacunación en función de la dinámica ganadera. Para evaluar mejor estos esquemas se desarrolló y aplicó un modelo estadístico de cuantificación de riesgos para asistir en la evaluación de planes de vacunación sistemática contra la fiebre aftosa de bovinos. El modelo utiliza información sobre la epidemiología de la fiebre aftosa; de la dinámica poblacional y de vacunación recolectada en un período llamado Ventana de Observación/Estimación y la proyecta hacia una fecha futura ubicada en un período llamado Ventana de Predicción en donde se obtuvo un predictor de la probabilidad de contacto entre terneros no vacunados y bovinos residualmente infectados por el virus de la fiebre aftosa. El modelo utiliza estimación de parámetros obtenidos de la opinión experta; de evidencias empíricas; o de la conjugación de ambos mediante estimadores bayesianos de las distribuciones Beta y Dirichlet. El modelo se aplicó a datos de Argentina mediante simulación de Monte Carlo, permitiendo identificar diferencias significativas al 5% entre los efectos de tres alternativas de vacunación comparadas mediante el método de Bonferroni.(AU)


The recurrence and persistence of fmd could be the consequence of cyclic and massive transportation of calves. For this reason, vaccination schemes related to livestock dynamic are promoted in South America. To improve the evaluation of vaccination schemes a quantitative stochastic risk assessment model was developed and applied in order to aid in the evaluation of strategies of systematic vaccination of cattle against fmd. The model uses information about fmd epidemiology and about population and vaccination dynamics. The information is collected during a period of time called Observation/Estimation Window and projected to a later time called Prediction Window where a predictor of the probability of contact between non-vaccinated calves with residually fmd infected cattle is obtained. Estimates of the parameters of the model are obtained from: expert opinion; empirical evidence or the conjugation of both by means of Bayesian estimators of the Beta and Dirichlet distributions. Applied to data of Argentina, through Monte Carlo simulation, the model allowed the identification of significant statistical differences among the effects of three different vaccination alternatives compared by Bonferroni test.(AU)

Na bovinocultura brasileira, a vacinação contra o vírus da Febre Aftosa (FA) é fundamental para a fase inicial de erradicação da enfermidade. Mesmo a qualidade da vacina tendo controle rígido feito pelos órgãos oficiais, restam variáveis técnicas ainda não monitoradas como manipulação, transporte e conservação pelo consumidor, dose, local e forma de aplicação que interferem na resposta imune, preocupações essas, que direcionam o presente estudo. Assim, pela pesquisa de anticorpos neutralizantes do vírus da FA em placas de microtitulação com cultivo de células BHK-21, foram determinados os títulos, calculados em logaritmo decimal (SN), em soros sanguíneos de bovinos vacinados conforme o protocolo apresentado. No primeiro grupo com 25 animais, a média de SN foi igual a 2,37 e 2,19, respectivamente, 30 e 180 dias após a vacinação, cuja vacina foi manejada por especialista com todos os cuidados técnicos recomendados. Outro grupo com 140 bovinos, distribuídos em 5 fazendas distintas, apresentou média de SN igual a 1,66 e 1,5l depois de 30 e 180 dias após a vacinação, cuja vacina foi manejada sem acompanhamento técnico e por indivíduos não especializados. Finalmente um terceiro grupo com 10 animais, que ficaram sem vacinação, apresentou média de SN igual a 0,82 e 0,81, também 30 e 180 dias após a aplicação do placebo. Assim, só os cuidados com a qualidade da vacina são insuficientes para proporcionar títulos satisfatórios que determinam proteção dos rebanhos contra o vírus da FA, uma vez que a literatura pertinente considera rebanhos com 1,52 de média do SN como tendo 50% dos animais protegidos, e com 1,70 como tendo mais de 70% de proteção, no período de até 7 meses.

ABSTRACT

Humoral immune response was evaluated by monitoring the serum antibody titres and virus specific IgM titres against Foot and Mouth Disease (FMD) virus antigens in serum samples obtained from different groups of calves inoculated with combined vaccine or FMD vaccine alone, on 0, 7, 14, 21, 28, 42 and 56 days post-vaccination (DPV). The cellular immune response was monitored by MTT based lymphoproliferation in peripheral blood mononuclear cell cultures. Higher liquid phase blocking (LPB) ELISA antibody titres were observed in calves receiving combined vaccine as compared to calves immunized with FMD vaccine alone with the peak titres in both the groups obtained on 21 days post-vaccination. However, the virus specific IgM titres were significantly higher in group of calves inoculated with combined vaccine than FMD vaccine alone. The lymphoproliferative responses against FMDV types O, A22 and Asia 1 in the groups receiving combined vaccine and FMD vaccine alone started increasing gradually after day 14 and reached peak levels on 28 DPV followed by a gradual decline subsequently. The group receiving combined vaccine showed higher proliferative responses on in vitro stimulation with FMD virus type O, whereas, with FMD virus type Asia 1, the responses were significantly higher on 14 and 21 DPV as compared to the group immunized with FMD vaccine alone. However, in the group receiving combined vaccine, the responses on in vitro stimulation with FMD virus type A22 were significantly higher than FMD vaccine alone group on all DPV except on 42 DPV.

ABSTRACT

Fifteen pregnant buffaloes at different periods of pregnancy and 5 non pregnant animals used as a control group were vaccinated with FMD vaccine and examined clinically for a week post-vaccination for any post-vaccinal reaction and the group of late stage of pregnancy was noticed after parturition for any stillbirths or abnormalities in their off springs. Serological analysis for serum neutralizing antibody titers were examined 15, 30 and 60 days post-vaccination for all animals. It is noticed that there were slight rise in the average body temperatures for 2 days postvaccination in the group of late stage of pregnancy and were normal in other groups. Also the average serum neutralizing antibody titers were lower in this group than in other groups especially few weeks before parturition. There were no abortion, stillbirths or any abnormalities in their off springs which denoted that FMD vaccine is safe for pregnant native breed buffaloes in Egypt

Duas vacinas foram submetidas a seis repetiçöes cada, à prova de eficiência, "Indice C", usando-se para titulaçäo, quatro cobaias por diluiçäo de vírus. Os valores encontrados, quando transformados em qualidade de vacina, demonstraram que uma mesma vacina poderia ser enquadrada como sendo "reprovada regular", "aprovada boa" ou "aprovada muito boa", indicando que a variaçäo dos resultados, dependendo do caso, pode indefinir a classificaçäo do imunógeno. Para verificar se tais variaçöes säo devidas ao pequeno número de cobaias, uma vacina foi submetida a seis repetiçöes, usando-se cinco cobaias por diluiçäo do vírus na prova "Indice C". Os arranjos 2 a 2, 3 a 3, 4 a 4, 5 a 5, realizados a partir dos resultados correspondentes a todas as combinaçöes possíveis quando säo usadas 5, 10, 15, 20, 25 ou 30 cobaias por diluiçäo viral, demonstraram que a variaçäo de mais ou menos 0,5 logaritmo com 95 por cento de segurança, corresponde a 15 cobaias

Foi desenvolvido um método de avaliaçäo de vacinas anti-aftosa em camundongos recém-nascidos, filhos de mäs vacinadas. Usou-se um índice, denominado índice proteçäo camundongos (IPC), resultante do quociente entre o título do vírus em camundongos filhos de mäes nä vacinadas (T/V). Verificou-se existir variaçäo nos resultados dos índices, e que para reduzí-la em níveis aceitáveis, é necessário usar, na titulaçäo, seis ou mais ninhadas de camundongos por diluiçäo de vírus
